No, they can't be certain whether a bid is placed via a sniping service.
Ebay does not provide a method for sellers to block bidders for that reason.
Sellers can of course see when bids are placed though and based on the timing might draw their own conclusions about how a bid was placed... but they can't be absolutely sure.
If they know the Ebay id where snipe bids are likely to be placed from they can block that, but that would be all bids, not just those placed in the last few seconds.
Also it's not possible for Ebay to force any seller to part with goods sold on their platform if they choose not to do so. They can cancel that sale after that auction has completed if they so wish... you would only really have the opportunity to leave negative factual and not profane (otherwise they might have grounds for removal) feedback on their account if they chose to do that.
If it were me I might use a sniping tool anyway and if challenged say I hadn't noted their small print in any case... but if you do win and they still want to refuse to sell to the highest bidder there's not a huge amount you can really do about it... if they did it too often I guess Ebay might consider blocking their account, but even that I think is unlikely.

I don't mean to be rhetorical but how can a seller make an assertion like this when eBay has stated sniping is not a policy violation? Since a seller can't actually tell if a sniping service was used this seems nuts for a seller to even "go there." Especially if the item is paid for before the seller decides they don't "like" the timing of your bid.
Seems it would drag down their seller rating if they keep canceling winning bids, buyers leave negative feedback - via web if they can or via phone support if they can't - and their feedback rating drops to whatever eBay considers unacceptable.
Besides, all a buyer would have to say is "no I didn't" if the seller asks if they used a sniping service. I'm not advocating dishonesty, but I can't abide stupid either.

P.S. I would like to point out that if the seller in this case refused to accept what appears to be a snipe at 6 seconds they would also have to accept the previous high bid - and lose out on over an additional $1,000.00 in the process.
Or perhaps this seller believes some snipes are better than others?

The last bid was 6 seconds before that auction end... with 4 other bids in the minute before that... so I guess we will see if the seller wants to sell at the price the auction was at a minute before the end... Like Rick I'm guessing they'll take the high bid.
